How do neurons instruct axon-dendritic polarity of synaptic receptors? Luís Ribeiro, Ben Verpoort, Joris de Wit and colleagues show that the presynaptic adhesion molecule neurexin reaches its destination via an indirect route after soma exit (image, cyan). Neurexin first traffics to the dendrite (image, magenta), where it is endocytosed, sorted by the endosomal sorting receptor SorCS1, and redirected to the axon (image, yellow). This sorting step in dendrites is essential for proper presynaptic development and function.
